Vilardell Glycerine Suppositories for Infants 0,92 G 15 Suppositories

Glycerol, the active ingredient in this medicine, is a laxative that is administered rectally. The laxative effect is achieved by the ability of glycerol to soften the stool which, together with a slight local irritant action, manages to stimulate bowel movements. It is indicated for the symptomatic relief of temporary and occasional constipation for infants and children under 2 years of age.

ACTION AND MECHANISM

- [LAXANT]. Glycerol is a polyol derivative that behaves as a laxative when administered rectally. Its effects are due to a double mechanism of action:

* Irritant effect on the rectal mucosa, without affecting the distal walls of the intestine. The local contraction of the rectal muscles causes the expulsion of feces in a short time.

* Osmotic effect. Glycerol has a great hygroscopic capacity, causing the passage of water towards the intestinal lumen. The increase in water, together with the lubricating properties of sorbitol, soften the stool and facilitate its elimination.

The effects take between 15-30 minutes and an hour to appear.

PHARMACOKINETICS

- Absorption: Glycerol is barely absorbed when administered rectally, being almost completely eliminated when defecation occurs. However, oral absorption is high. Maximum oral plasma concentrations are reached within 60-90 minutes.

- Distribution: The absorbed glycerol is widely distributed throughout the body, accumulating in muscle fat.

- Metabolism: It is extensively metabolized in the liver (around 80% of the amount absorbed).

- Elimination: Glycerol is eliminated mainly by hepatic metabolism. In urine, 20% of the absorbed content appears unchanged. The elimination half-life is 30-45 minutes.

INDICATIONS

- [CONSTIPATION]. Symptomatic local treatment of temporary and occasional constipation.

POSOLOGY

- Children and adolescents under 18 years of age, rectally:
* Children under 2 years: 1 suppository / 24 h.
