ACTION AND MECHANISM

- [LAXANT]. Glycerol is a polyol derivative that behaves as a laxative when administered rectally. Its effects are due to a double mechanism of action:

 

* Irritant effect on the rectal mucosa, without affecting the distal walls of the intestine. The local contraction of the rectum muscles causes the expulsion of feces in a short time.

 

* Osmotic effect. Glycerol has a great hygroscopic capacity, causing the passage of water into the intestinal lumen. The increase in water, together with the lubricating properties of sorbitol, soften the stool and facilitate its elimination.

 

The effects take between 15-30 minutes and an hour to appear.

PHARMACOKINETICS

- Absorption: Glycerol is barely absorbed when administered rectally, being almost completely eliminated when defecation occurs. However, oral absorption is high. Maximum oral plasma concentrations are reached within 60-90 minutes.

 

- Distribution: The absorbed glycerol is distributed widely throughout the body, accumulating in muscle fat.

 

- Metabolism: It is widely metabolized in the liver (about 80% of the amount absorbed).

 

- Elimination: Glycerol is eliminated mainly by hepatic metabolism. 20% of the absorbed content appears in unchanged form in urine. The elimination half-life is 30-45 minutes.

INDICATIONS

- Local symptomatic treatment of transient and occasional [CONSTIPATION].

POSOLOGY

- Adults, rectally: 1 suppository/24 hours (this is the general rule, although they can be administered when necessary, or according to medical criteria) or 1 application (adult rectal solution)/24 hours.


- Children and adolescents under 18 years of age, rectally:
* Children and adolescents over 12 years of age: 1 suppository/24 hours or 1 application (children's rectal solution)/24 hours.
It is recommended not to use for periods longer than 6-7 days, unless medically advised. If after this time the symptoms continue or worsen, it is advisable to consult a doctor and/or pharmacist.

 

 

RULES FOR CORRECT ADMINISTRATION

- Suppositories: It is recommended to previously harden the suppositories by placing them for 30 seconds under cold tap water or 10 seconds in a glass of water. The suppositories will be inserted deep into the anal opening.

Repress evacuation for as long as possible so that the medication can exert its action perfectly.

 

ADVICE TO THE PATIENT

- Before starting treatment for constipation, lifestyle habits should be modified. An adequate daily intake of fluids and fiber, responding quickly to the stimulus of defecation, and performing physical exercise are recommended.

- This medication should not be used by patients with intestinal obstructions.

 

- Consult your doctor and/or pharmacist if constipation worsens, or if symptoms such as nausea or vomiting, abdominal bloating or fever appear.

 

CONTRAINDICATIONS

- Hypersensitivity to any component of the medication.

- Situations that could be aggravated by the laxative effect, such as [INTESTINAL OBSTRUCTION], [INTESTINAL PERFORATION], chronic [INFLAMMATORY BOWEL DISEASE], [APENDICITIS] or [TOXIC MEGACOLON]. Logically, it should not be used in patients with [DIARREA].

 

PRECAUTIONS

- Symptoms of intestinal obstruction. Before starting treatment with a laxative, it is advisable to perform a differential diagnosis of intestinal obstruction in patients with symptoms such as [NAUSEA], [VOMITING] or [ABDOMINAL PAIN] of unknown origin, or [ABDOMINAL DISTENSION].

 

SPECIAL WARNINGS

- Perform a differential diagnosis in patients suspected of intestinal obstruction.

 

INTERACTIONS

PREGNANCY

Safety in animals : no data available.

Safety in humans : There have been no adequate and well-controlled studies in humans. This medication will be used according to medical discretion.

Effects on fertility : No specific studies have been performed in humans.

 

LACTATION

Safety in animals: no data available.

Safety in humans: it is unknown if it is excreted with milk, and the consequences it could have for the infant. It will be used according to medical criteria.

 

CHILDREN

Its use in children under 12 years of age is not recommended.

ADVANCED AGE

No specific problems have been described in this age group, although the chronic use of laxatives in elderly patients can trigger phenomena of orthostatic hypotension, weakness and motor incoordination due to loss of fluid and electrolytes.

ADVERSE REACTIONS

Adverse reactions from glycerol are generally mild and transient, and local in nature.

 

- Digestive. Occasionally irritation, stinging or [ANAL ITCHING] may appear.

OVERDOSE

Symptoms: Using this medication for long periods of time and at high doses may lead to irritable bowel syndrome.

 

Treatment: Treatment will be symptomatic. Administration of the laxative should be discontinued.
