CHRONIC VENOUS DISEASE

Around 60% of the adult population worldwide suffers from Chronic Venous Disease (EVC). Under normal conditions, the veins lead the blood back to the heart from the legs. For this they use valves that facilitate the flow of blood in the opposite direction to the force of gravity. Venous insufficiency (IV) is a condition in which the veins in your legs have difficulty returning blood to your heart. This pathology affects the valves of your veins whose main function is to prevent venous reflux. Problems with these valves cause a slowing of the venous return flow that causes blood to accumulate pathologically in the veins of the legs.

WHAT IS DAFLON 500?

Daflon 500 is indicated for the treatment of symptoms related to mild venous insufficiency in adults. Relieves, in adults, the symptoms of mild venous insufficiency of the legs, such as: pain, feeling of heaviness, tightness, tingling and itching in the legs with varicose veins and swollen legs.

TREATMENT

The usual therapeutic dose is 2 tablets per day, divided into two doses, one tablet at noon and one at night, with meals.

WHY DAFLON® 500?

It has an exclusive composition, with proven synergistic efficacy of its purified micronized flavonoid fraction (FFPM). 
Its patented micronization reduces the particle diameter from 36.5 µm to 1.7 µm, which favors the absorption of the drug.

It can prevent the evolution of the disease and generates greater improvement of the symptoms as the treatment progresses.

ACTION AND MECHANISM

Daflon 500 is a venotonic and vasoprotector. It produces venoconstriction, increased resistance of the vessels and decreased permeability.

In the animal, it exerts a double action on the venous return system:

- at the level of veins and venules: it increases parietal tonicity and exerts an antistatic action;

- at the level of microcirculation: normalizes capillary permeability and strengthens capillary resistance.

In man, double-blind controlled studies have confirmed venotonic activity: venous tone increases: plethysmography of venous occlusion with a mercury ring has revealed a decrease in venous emptying times.

It has also shown activity on microcirculation: in patients who show signs of capillary fragility, capillary resistance measured by angio-sterometry increases.

PHARMACKINETICS

The pharmacokinetics of diosmin (daphlon) is linear.

Orally:

- Absorption: Daflon 500 is rapidly transformed by the intestinal flora and is absorbed in its aglycone form (diosmetin). Bioavailability: 57.9% approx.

- Distribution: extensive (Vd: 62.1 L).

- Metabolism: Diosmetin degrades rapidly and widely to phlebotonic acids or their derivatives conjugated with glycine, which are eliminated in the urine. Predominant metabolite: hydroxyphenylpropionic acid (which is mostly eliminated in conjugate). Metabolites in smaller amounts: other phenolic acids corresponding to 1-hydroxy-4-methoxybenzoic acid, 3-methoxy-4-hydroxyphenylacetic acid and 3,4-dihydroxybenzoic acid.

- Elimination: relatively fast. In studies with radioactively labeled diosmin, 34% of the dose was recovered in urine and feces after the first 24 hours, and approximately 86% recovered in urine and feces after the first 48 hours.

DAFLON 500 INDICATIONS

- Daflon 500 provides short-term relief (2 to 3 months) of edema and symptoms related to chronic [VENOUS INSUFFICIENCY].

DAFLON POSOLOGY

- Adults: Usual therapeutic dose: 500 mg / 12 h at noon and at night, with meals.

- Children and adolescents <18 years: not recommended.

- Elderly: no dose adjustment required.

Duration of treatment : 2-3 months maximum.

RULES FOR THE CORRECT ADMINISTRATION OF DAFLON 500

Take Daflon 500 with meals (at noon and dinner).

CONTRAINDICATIONS OF DAFLON 500

- Hypersensitivity to diosmin or other flavonoids.

ADVICE TO THE PATIENT WHO TAKES DAFLON 500

- It is recommended to take the tablets after lunch and dinner.

- This medicine is intended for short-term treatment of venous insufficiency, for periods of less than 2-3 months.

IT IS SAFE TO TAKE DAFLON 500 IN PREGNANCY

There is no record of any harmful effects on the human species. Experimental studies have not shown any teratogenic effect.

Extreme caution should be exercised in prescribing pregnant women.

IT IS SAFE TO TAKE DAFLON 500 DURING BREASTFEEDING

It is not known if this medicine is excreted with breast milk, however, in a study in animal models there has been an excretion in minimal breast milk (1%). In the absence of conclusive data on the transition to breast milk, its use is not recommended during breastfeeding.

DAFLON 500 IN CHILDREN

There is not enough clinical experience in this age group. Not recommended use.

DAFLON 500 IN ELDERS

In one study there were no significant differences with respect to adverse effects between geriatric patients and younger patients. Accepted use.

DAFLON 500 ADVERSE REACTIONS

Adverse effects of diosmin are generally rare, mild and transient.

Adverse reactions are described according to each frequency range, being considered very frequent (> 10%), frequent (1-10%), rare (0.1-1%), rare (0.01-0.1%) , very rare (<0.01%) or of unknown frequency (cannot be estimated from the available data).

No cases of overdose have been described. The wide therapeutic margin of this specialty determines that the risk of intoxication is practically nil.
